N N O O T T E E : : For Serum Screen, 1 sample application is required. For Urine Screen, 10 sample
applications are required. Remove the gel blocks prior to drying.
8. Following electrophoresis, ( ( S S A A S S - - 1 1 P P l l u u s s u u s s e e r r s s : : remove the cover), remove the electrodes from the
surface of the gel. ( ( S S A A S S - - 3 3 u u s s e e r r s s : : remove the alignment guide). Position the antiserum application
template onto the gel surface. NOTE: The milled antisera channels should be aligned centrally
over the printed box on the gel in which the samples are applied.
9. Apply 60µL of Pentavalent Antiserum into each hole of the antiserum application template. Ensure
that the antisera has completely filled the channels.
10. Incubate the gel.
11. Following incubation, place a blotter comb into the holes of the antiserum template. Allow 2
minutes for the excess antisera to be absorbed, then remove the blotter combs and the template
from the gel surface.
12. Place a blotter D (smooth side down) onto the surface of the gel, leave for 10 seconds and remove.
13. Place a blotter D (smooth side down) onto the surface of the gel and replace the antiserum
template to hold the blotter flat. Blot the gel. NOTE: Immediately after use, clean the antisera
template with a mild biocidal detergent.
14. Remove the blotter D. S S A A S S - - 3 3 u u s s e e r r s s : : Dry the gel, and start initial 3 second chamber wash on
SAS-4 unit.
15. Remove the gel blocks and attach the gel to the staining chamber holder, and complete the final
steps on the SAS-2 or SAS-4.
16. At the end of the staining cycle, remove the gel from the staining chamber. The gel is now ready
for examination.
