PROBLEM
Transfer blot is uneven
- The blotting membrane must make full, even con-
tact with the gel in the blot stack.
- There should be no bubbles between the different
layers of filter paper, gel or membrane in the blot stack.
- The gel must be fully equilibrated in transfer buffer.
Sample detection is missing
- Verify the proper order of the gel and the membra-
or weak
ne in the stack.
- For larger molecules, increase the transfer time.
- Decrease the acrylamide or biscarylamide concen-
tration of the gel to increase large molecule transfer
mobility.
- Do not transfer molecules for as long or used higher
acrylamide concentration.
- The wrong type of membrane was used.
- Add some SDS to the transfer buffer to ensure that
the proteins have a negative net charge.
- For small proteins, use smaller (0.22 m) pore mem-
branes.
- Include a lane of prestained protein molecular
weight standards for a visual check of the transfer pro-
cedure.
To get the best results and a higher duration of the horizontal electrophoresis cell it
is essential to follow the processes of use.
Note: All the processes of use mentioned below will not have any value unless you keep
a continued and careful maintenance.

This manual should be available for all users of this equipment.
Always use original components and supplies. Other devices can be similar but they
can damage the equipment.
Never autoclave or dry-heat sterilize the apparatus or components.
Do not expose the apparatus or components to phenol, benzene, acetone, halogenated
hydrocarbon solvents or undiluted laboratory alcohols.

1. APLICACIONES DEL INSTRUMENTO
El módulo de transferencia Nahita está diseñado para ser utilizado junto con el tanque
externo y la tapa de la cubeta de electroforesis vertical Nahita para la transferencia de
moléculas (proteínas o ácidos nucleicos) desde el gel en el que han sido separados hasta
una membrana para su posterior detección. Para ello, el equipo dispone de dos casetes de
transferencia para acomodar cada uno de los geles de 82x82 mm generados en la electro-
foresis vertical realizada previamente.
Las diversas características del módulo de transferencia lo convierten en un equipo fácil
de manejar, seguro para el usuario y de una gran eficacia para la transferencia molecular.
