Componentes
El módulo de transferencia Nahita es totalmente compatible con el tanque externo y la
tapa de la cubeta de electroforesis vertical Nahita y consta de los siguientes componentes:
- Soporte para transferencia: fabricado en material de
alta calidad con una gran resistencia a la corrosión.
Presenta dos electrodos de platino puro resistentes a
la corrosión y a elevadas temperaturas que se
encuentran paralelos a una distancia de 4.5 cm. La
fuerza direccional para la transferencia de las molé-
culas viene dada por el voltaje generado a lo largo de
la distancia entre ambos electrodos. En el caso del
soporte para transferencia, esta pequeña distancia
permite la generación de elevadas fuerzas direccio-
nales lo que asegura una transferencia muy eficiente
de las proteínas. El soporte además presenta un lado
de color rojo y el otro negro para asegurar una correc-
ta orientación del gel durante el proceso de transfe-
rencia.
- Casetes con placas perforadas: el equipo incluye dos
casetes para alojar cada uno de los geles obtenidos
durante el proceso de electroforesis con la cubeta de
electroforesis vertical de Nahita. Los casetes están
fabricados en policarbonato de gran resistencia a la
rotura, la acción de agentes químicos y a presión y
temperatura elevadas. Una de las placas de cada case-
te es de color negro lo que permite orientar correcta-
mente el casete con el gel dentro del soporte para
transferencia. Además, cada casete presenta una pinza
de cierre para sujetar firmemente el montaje.
- Almohadillas: necesarias para el montaje de transferencia.

- Put the cassette into one of the compartments of the transfer support so as the
black plate faces the black side of the support and the latch stays upwards.
- Follow the same protocol to make the stack for the transfer of the second gel.
- Put the transfer support with the cassettes and the stacks into the tank of the
vertical electrophoresis cell and fill it with approximately 500 mL of transfer buffer.
- Cover the tank with the lid so as the red lead is connected to the red electrode
(positive pole, anode) and the black lead to the black electrode (negative pole, cathode).
- Plug the power leads to a power supply
- Switch the power supply on and set the parameters of voltage and current nee-
ded for transfer blot. Usually parameter setting is determined by the size of the membra-
ne. E.g. if the membrane is 7x6 cm, the constant current should be 7x6x2=84 mA for a
transfer period of 2 h.
After transfer blot
- When transfer is complete, switch the power supply off and unplug the power
leads. Remove the lid from the tank.
- Take out the transfer support and properly discard the transfer buffer. Do not
reuse the buffer.
- Take out and open each cassette. Using gloved hands and forceps remove the
pad, discard the filter paper and take the membrane. Process the membrane as desired.
- After each use, rinse all components, including pads, with deionized water.
Security
The transfer module must be used by previously qualified staff that knows how the
equipment works thanks to the user manual.
Always work on top of a horizontal, plane and stable table.
Do not put the transfer module near any warm supply (burners, blowlamps...). Avoid
vibrations.
The transfer module is intended to use with the tank and the lid of the Nahita vertical
electrophoresis cell (code 53001001). The lid is then plugged to a DC power supply. An
inadequate manipulation may cause burns, hurts and damages to the electrical installa-
tion.
Never connect the lid of the electrophoresis cell to the power supply before starting the
experiment. The electrical current supplied by the power supply is transmitted to the cell
through the leads and the lid assembly.
Never use the transfer module and the tank of the electrophoresis cell without the lid.
Always turn off the power supply before opening and removing the lid.
Never operate damaged or leaking equipment.
Certain reagents indicated for use in this manual are of a hazardous nature (e.g. metha-
nol). User should strictly observe the safety regulations indicated for such products.
The equipments used in these procedures (e.g. power supplies) should be used follo-
wing the manufacturer safety recommendations.
