solutions that contain undissolved components (suspensions) – in particu-
lar nutrient media with Agar and/or other jellying agents; if possible avoid
the strong stratification of solutions and mix the contents up intensively
just before inserting into the process chamber.
residues precipitating at the bottom may obstruct the even heating of the
sample – especially when they are abundant. Practical experience showed
that suspensions containing more than 12 g/l of undissolved substance
require special care and mixing up just before they are inserted into the
chamber.
agar medium: Powder or granular media containing agar are mixed
with water at room temperature to completely dissolve the powder or
granules. it is forbidden to use not dissolved granules. after mixing, flask
should be put in the work chamber device as soon as possible and than
process should start. When agar sink to the bottom of the vessel, re-mixed
is required to obtain a homogeneous solution before placing vessel in the
working chamber and performing sterilization. For mixing the powder or
granular substrates may be used a magnetic stirrer. it's allowed to leave di-
pole in the vessel during sterilization. after the sterilization process should
be repeated stirring process, especially when we work with Petri dishes.
Make sure to use only Teflon (PTEE) coated stirrers– only these are
resistant to high temperature!
