4.0 USING THE MICROSCOPE
4.14 Using the Phase Contrast set (optional)
The phase contrast set has 4 objectives for phase contrast and one condenser, with 4 rings of
phase, and one position for brightfield. To use the phase contrast, remove the Abbe condenser of the
microscope and replace it with the phase contrast condenser.
If, during use, the phase contrast diaphragm rings (anuli) are dislodged, they can be realigned rotating
the alignment screws placed on two sides of each ring, using the enclosed tools. In particular:
-Select an objective corresponding to the ring to be realigned, and focus on a slide under brightfield
illumination.
- The selector of the condenser has 5 selectable positions:
0 → to use when it is necessary to be used the instrument in brightfield
10 → to use with 10x phase contrast objective
20 → to use with 20x phase contrast objective
40 → to use with 40x phase contrast objective
100 → to use with 100x phase contrast objective
- Insert the phase ring to be realigned.
- Remove an eyepiece, and replace it with the centring telescope.
- Focus the centring telescope, without touching the microscope focus, in order to focus the phase ring
and the phase plate simultaneously. In particular, the phase plate is visible as a dark ring, and the
phase ring as a bright ring divided into sectors.
- Turning the two phase ring centring screws, position the phase ring (bright ring) so that it is
completely contained in the phase plate profile (dark ring). (See figure.)
- Remove the telescope, and replace the eyepiece.
4.15 Darkfield condenser (optional)
For darkfield observation, the Abbe condenser in standard equipment must be replaced with a special
condenser for darkfield, showed in the picture.
