3.2 Filtration Process
The filtration process can be performed when the cassette is connected to a
peristaltic pump. Macromolecules sufficiently larger than the nominal pore
size of the membrane are retained in the flow path and recirculated between
a feed reservoir and the cassette, while solvent and microsolutes permeate
the membrane. This process results in progressive concentration of the mac-
romolecule of interest. With the addition of a suitable exchange buffer, the
product can also be used in diafiltration mode for sample desalting or buffer
exchange.
