The HistoCore SPECTRA H&E Staining System S2 contains 5 proprietary reagent components and staining protocols that when used together provide
consistent diagnostic quality and ease of use. This system has been optimized for use on HistoCore SPECTRA ST stainers.
Intended Use
The reagents in this HistoCore SPECTRA H&E Staining System S2 are intended for in vitro diagnostics use only. The reagents in the HistoCore
SPECTRA H&E Staining System S2 are intended for use on HistoCore SPECTRA ST Stainers. The HistoCore SPECTRA H&E Staining System S2 is
used to stain histological samples with Hematoxylin and Eosin for subsequent microscopic evaluation.

Storage and Stability
Store reagents at room temperature (15-30°C). Please follow local storage requirements for flammable products.
CAUTION: Do not use after the expiration date.
Specimen Preparations
Fixation
Any general fixative including but not limited to neutral buffered formalin may be used. NOTE: Under fixation or over fixation may result in
compromised staining quality.
Paraffin Sections
Following processing and paraffin embedding, section tissues at standard thickness (2 – 5µm).
Directions for use of the HistoCore SPECTRA H&E Staining System S2:
Settings for operational parameters of the HistoCore SPECTRA ST stainer and protocol schedules are provided to optimize the performance and
consistency of the HistoCore SPECTRA H&E Staining System S2.

Slides and Specimens
If the oven step is not being utilized on the instrument prior to staining, slides should be dried at 66 - 74°C for 15-22 minutes.
Oven Specifications:
To either activate the oven step, or deactivate the oven step for the SPECTRA H&E protocol, refer to the HistoCore SPECTRA ST Instructions for
Use.
Placing the HistoCore SPECTRA Staining System S2 on the Instrument:
Scan the HistoCore SPECTRA H&E staining system box and each individual bottle at the Radio Frequency Identification (RFID) scanner located on
the left side of the HistoCore SPECTRA ST stainer. Follow the prompts as indicated by the instrument to place the reagents into the designated
reagent vessels.
HistoCore SPECTRA ST Bathlayout
When prompted by the user, the bathlayout is automatically generated by the instrument. The bathlayout includes specific protocols chosen by the
user. The instrument will assign each reagent to a specific reagent station. The 95% alcohol in step 16 of the protocol must be duplicated manually
as described below:
In the station assignment view, press the "Adapt Bathlayout" button.
To duplicate a reagent station, select it in the bathlayout by touching the screen.
Finally, press the "Duplicate" button. Then, the reagent station will appear in the clipboard with an equivalence symbol (∞).
After this, select the duplicate reagent station in the clipboard by touching the screen and place it in the desired (available) station, near
the original station in the bathlayout.
To save your changes, press the "Finish" button and confirm the subsequent message with a "Yes" or "No" to return to the clipboard.

Agitation Settings:
Agitation settings are predefined. Agitation will occur at a setting of 4. Refer to the HistoCore SPECTRA ST Instructions for Use.
Immersion Times:
Immersion times and instrument settings for HistoCore SPECTRA ST stainer are detailed in Table 1 below. Please follow instructions for operating
HistoCore SPECTRA ST as detailed in the HistoCore SPECTRA ST Instructions for Use.
CAUTION: Modifications to the validated staining protocol may result in compromised staining quality as well as staining capacity.
CAUTION: All of the reagents in each HistoCore SPECTRA H&E Staining System S2 must be used together to achieve the expected performance.
Substitution of non- HistoCore SPECTRA H&E reagents or reagents from another HistoCore SPECTRA H&E Staining System will compromise
performance.
CAUTION: Opening the lid of the HistoCore SPECTRA ST stainer during protocol execution may increase time in staining system reagent and impact
the consistency in staining quality.
CAUTION: Reagent vessels should be covered when not in use to reduce evaporation.
