CAUTION: If the HistoCore SPECTRA ST is not connected to the HistoCore SPECTRA CV, the slides will be transferred to the unload drawers. In this
occurrence, ensure there is the appropriate reagent in the unload vessel, otherwise quality of staining may be compromised.
CAUTION: Any deviations from these instructions may alter expected results and are neither validated nor supported by Leica Biosystems.
Technical Notes
100% alcohol and xylene reagents used for deparaffinization and rehydration should be rotated after every 800 slides. Reagent rotation is defined
as follows:
Empty most contaminated reagent of a series and refill container.
Move remaining container(s) up one space in sequence.
Place newly filled container into the last position of the series.
The 100% alcohol and xylene reagents used for dehydration and clearing before coverslipping also should be rotated after every 800 slides as
described above.
Two of the four 95% alcohols (steps 5 and 14) should be discarded and replaced with fresh 95% alcohol solution every 800 slides.
The two 95% dehydrating alcohols (step 16) should be discarded and replaced with fresh 95% alcohol solution every 400 slides.
If using the Reagent Management System (RMS), after change and rotation, update the RMS with changes. Instruction for use of the RMS can be
found in the HistoCore SPECTRA ST Instructions for Use.
CAUTION: Failure to rotate and change reagents as required may result in inadequate deparaffinization as well as excessive carry-over and dilution
of reagents or solvent.
NOTE: Reagent vessels (including water rinse vessels) should be inspected at periodic intervals to insure the integrity and cleanliness of the
vessels. If cleaning solutions are used, rinse vessels with adequate water to remove any residual cleaning solutions prior to use. Clean the vessels
in accordance with the instructions provided in the HistoCore SPECTRA ST Instructions for Use.
NOTE: The HistoCore SPECTRA ST Reagent Management System (RMS) tracks the number of slides that have utilized a specific station.
However, for the ancillary reagents that have been rotated, the RMS will no longer capture the total number of slides that have utilized that
specific ancillary reagent. For more information please refer to the SPECTRA ST Instructions for Use.
