Defining a Bathlayout
Single S1/S2 Staining System:
To use the HistoCore SPECTRA H&E Staining System S1/S2, a bathlayout must be generated. Follow the steps below to generate
a bathlayout containing the HistoCore SPECTRA H&E Staining System S1/S2 protocol:
•
Under the "Program" tab, press the button "Define Bathlayout" located at the bottom of the screen.
•
A new window will open. In this window, select the box under the "Use" column next to the corresponding staining
system protocol desired: "SPECTRA H&E S1 A" or "SPECTRA H&E S2 A".
NOTE: If additional protocols wish to be generated, they must be selected at this time.
Once all protocols have been selected, press the button "Execute Bathlayout" at the bottom right hand of the screen.
•
A message will appear containing information about the need to use fresh reagents. Press "Continue" to proceed,
or "Cancel" to abort bathlayout execution.
CAUTION: Once the "Continue" button is pressed, any and all staining system(s) and reagents will be erased from the
bathlayout, regardless of the current state of the staining system(s) or reagents. This will make any staining system(s)
and reagents unusable.
•
Once "Continue" has been pressed, a new window will open. This window will ask you to confirm the protocols
selected to be adapted in the bathlayout. Press "Continue" to proceed.
•
A screen demonstrating the bathlayout will open. A message will appear at the top of screen stating, "For validated HistoCore
SPECTRA H&E program(s) the 95% Alcohol Dehyd 1 (95Dhy 1) must be duplicated using the function "Adapt Bathlayout"."
To complete this step, press the "Adapt Bathlayout" button. An additional window will open on the screen. This window
is called a "Clipboard" window. An identical copy of the selected station will appear on the "Clipboard". Select the station
in the "Clipboard" and then click on a free position in the bathlayout. The free station selected should be as close to
the SPECTRA Eosin S1/S2 (L Eos S1 or L Eos S2) and 100% Alcohol Dhyd 1 H (100Dhy 1H) as possible. The duplicated
station has now been moved from the clipboard to the bathlayout. Press the "Finish" button on the "Clipboard" and
confirm the subsequent message with "Yes" in order to apply the change and close the clipboard. Both 95Dhy 1 stations
will now display an equivalence symbol (∞) in the bathlayout.
Dual S1 or S2 Staining Systems:
To use dual HistoCore SPECTRA H&E Staining System S1/S2, a bathlayout must be generated. Follow the steps below to
generate a bathlayout containing a HistoCore SPECTRA H&E Staining System S1/S2 protocol with two H&E staining lines:

Under the "Program" tab, press the button "Define Bathlayout" located at the bottom of the screen.

A new window will open. In this window, select the box under the "Use" column next to the corresponding staining
system protocol desired: "SPECTRA H&E S1 A" and "SPECTRA H&E S1 B" or "SPECTRA H&E S2 A" and
"SPECTRA H&E S2 B".
NOTE: If additional protocols wish to be generated, they must be selected at this time.
Once all protocols have been selected, press the button "Execute Bathlayout" at the bottom right hand of the screen.
•
A message will appear containing information about the need to use fresh reagents. Press "Continue" to proceed,

SPECTRA H&E S2 A (S2A), SPECTRA H&E S2 B (S2B) and any additional protocols to be generated, press "Continue"
and proceed.

A screen demonstrating the bathlayout will open. A message will appear at the top of screen stating, "For validated
HistoCore SPECTRA H&E program(s) the 95% Alcohol Dehyd 1 (95Dhy 1) must be duplicated using the function Adapt
Bathlayout." To complete this step, press the "Adapt Bathlayout" button at the bottom of the screen. An additional window will
open on the screen (Clipboard). Select the 95Dhy 1 station in the bathlayout. Select the "Duplicate" button on the Clipboard.
The duplicated station will be placed onto the "Clipboard". Both 95Dhy 1 stations will display an equivalence symbol (∞).
NOTE: To operate dual HistoCore SPECTRA H&E Staining System S1/S2 at the highest efficiency, it is recommended to
duplicate all ancillary reagent stations, and quadruple the 95Dhy 1 station. To duplicate all ancillary reagent stations,
follow the steps listed above. A suggested bathlayout with dual ancillary reagents and dual staining lines is demonstrated
in Image 1 below.
